Midfielder Yegor Titov tested positive for Bromantan after the 0-0 play-off first leg with Wales on 15 November.
Russia won the return leg in Cardiff 1-0 four days later, so Wales will approach Uefa on Monday to call for the result to be declared void.
But on Saturday, a Uefa spokesman said it was "highly unlikely" that it would be grounds for altering the result.
Wales manager Mark Hughes has been unhappy after previous dealings with European football's governing body and is also doubtful whether any appeal would succeed this time.
"The worry with Uefa is that in the past they haven't really wanted to push things where Wales are concerned, so I don't know how far we can go with it," Hughes said.
"I think the situation we are in now we have to really pursue our options and see whether Uefa are going to do something about it."
That seems unlikely as Uefa spokesman Rob Faulkner stated that: "It is highly unlikely that any such complaint from the FA of Wales would have any grounds for altering the result of the Russia-Wales match."
Although Hughes may not be hopeful of the outcome, he is determined that the Football Association of Wales should appeal.
"My initial reaction was shock because it's not something you expect to hear about," Hughes added.
"Obviously when it impacts on the result against the (Russia) side we were involved with it begs questions."
Bromantan has been called the "secret" Russian drug and positive tests led to Russian athletes being stripped of their medals at the 1996 Olympic Games in Atlanta.
They were reinstated because the then-unknown substance had not been on the International Olympic Committee's banned lost.
It is a stimulant and masking agent and is highly likely to have still been in the player's system for the second leg.
"In the double-header games there is an impact on energy levels in the second game," Hughes said.
"You have to say looking back on the game in Cardiff, they certainly finished stronger than we did.
"I'm not trying to suggest anything more, but unfortunately when stories like this break it makes people question things."
The Football Union of Russia announced it would not be appealing against Titov's 12-month ban and �4,420 fine, with his club Spartak Moscow also being hit with a fine of �8,840.
However, Titov is expected to appeal the ban with the support of his club.
Spartak say Titov was prescribed the medication Remontadin by a team doctor to cure a cold, with club officals believe may have contained traces of Bromantan or a similar substance.
The Spartak skipper was an unused substitute in Moscow, but played 59 minutes before being substituted in the second leg at the Millennium Stadium. |
