Welllll......tall order. I'll do my best.
Terok Nor was a Cardassian space station built in orbit of the planet Bajor during the Cardassians' occupation of that world. Eventually, the Cardassians were driven away by the Bajoran resistence. At that point, the new Bajoran provisional government decided they needed allies in case the Cardassians returned, and asked the Federation for help.
The Federation sent officers to take charge of Terok Nor, which they renamed "Deep Space Nine". These included Commander Benjamin Sisko, who was on the USS Saratoga at the famous battle of Wolf 359 (when Picard was a Borg). Sisko's wife had been killed in that battle, but his son survived.
The other officers are CMO Julian Bashir, who is genetically engineered (though we don't find out until season 5), Jadzia Dax, a Trill, Kira Nyres, a former member of the Barjoran Resisitence, Miles O'Brien, previously assigned to the Enterprise-D, and Odo, a shapeshifter raised by a Bajoran. Also on the station are three Ferengi---Quark, owner of the station's bar, his brother, Rom, and nephew, Nog. Garak, a Cardassian exiled from his homeworld, runs a tailor shop.
Very soon after Starfleet took over the station, they discovered a wormhole leading to the Gamma Quadrant nearby. Using the Station's few working thrusters, they break orbit of Bajor and move the station the mouth of the wormhole.
Sisko discovers there are beings living in the wormhole----the "Prophets" worshiped by the Bajorans. Since he was the first to make contact with them, Sisko is declared the Emissary of the Prophets----a title he is at first uncomfortable with, but learns to accept.
Unbeknownst to Starfleet, the Gamma Quadrant isn't just another place to explore. It is the home of the Dominion. They're bad guys, basically. Tensions mount between the Federation and Domionion, until at last war breaks out in the season 5 finale.
Commander Worf, formerly of the USS Enterprise, joins the crew for season 4.
I think that's all you really need to know at the moment. One thing to remember is that DS9 is a show you *really* need to see from the beginning, in order. The first two seasons are only so-so in quality, but they set up important backstory for the later years (which are quite good).
A few of the highlight episodes:
Civil Defense
Way of the Warrior (2 hr)
The Visitor
Call To Arms (through Sacrifice of Angels)

Oh, yeah-----a word on the villians of the show.
Gul Dukat was the former administrator of Terok Nor. He's somewhat bitter about losing his post. However, while he's *ultimately* a bad guy, there are times when he allies himself with the crew if it serves his purposes.
Weyoun is a Vorta. Vorta are servants of the Domionion, cloned beings designed to be intelligent and to lead the Dominion's shock troops, the Jem'Hadar (who are also genetically engineered). Weyoun is first seen in the episode "To The Death", but he quickly becomes a major bad guy. He's like a used car salesman, only worse. He's played by the same guy as Shran the Andorian in Enterprise. In fact, my avatar is of him at the moment.
DS9 deals with secret organizations a lot. Whether it be the Romulan Tal Shiar, the Cardassian Obsidian Order, or even the Federation's own Section 31....well, its pretty cool that way.
Oh, you asked about the Cardassians? They've been in both TNG and VOY a couple of times. Hmmm....did you see the VOY episode "Flesh and Blood"? The engineer hologram was a Cardassian.
Hmmm.....you know the Maquis from Voyager? They originated in DS9. They aren't major villians---in fact, its hard to call them villians at all---but they are a recurring presence in the series, until they are wiped out by the Dominion in the episode "Blaze of Glory". It was the events of that episode which Chakotay finally hears about in the VOY episode "Hunters".

The Trill are first seen in the TNG episode "The Host". However, DON'T go to that episode for information about them, because virtually everything about them except the basic premise is re-imagined in DS9.
That premise is that they are a joined race. The hosts can carry Symbionts within them. Jadzia is the seventh host of the Dax symbiont; Ezri (season 7) is the eighth. Not all Trill are joined; to be joined is considered an honor.

Curzon was shown in "Emissary", briefly. Then, in the episode "Faces", the other memebers of the crew briefly take on the personalities of the past hosts, but you don't see what they look like.
Well.....exect when Odo takes on Curzon's personality. That has some......interesting results.

Ack...I gotta find a way to see those episodes!
